Unexpected Surge: Moderna Jumps 177% on Historic Cancer Vaccine Breakthrough, Sending ASX and Hong Kong Biotech Stocks Soaring

In a market move that has caught many by surprise, biotech giant Moderna experienced a staggering single-day surge of 177% this week, following the release of highly anticipated clinical trial data. The announcement has created a powerful ripple effect, igniting a wave of limit-up trading across innovative drug and vaccine stocks on both the mainland A-share and Hong Kong markets.

On August 19, pharmaceutical heavyweight Merck and Moderna unveiled positive topline results from a pivotal Phase III clinical trial. The study, which enrolled 1,137 patients with high-risk melanoma, evaluated the efficacy of their jointly developed personalized mRNA cancer vaccine, known as intismeran autogene (V940 / mRNA-4157). The trial design compared the combination of intismeran autogene with pembrolizumab against pembrolizumab monotherapy. The results successfully met the primary endpoint of recurrence-free survival (RFS) and the key secondary endpoint of distant metastasis-free survival (DMFS). The research will continue, with analysis of the overall survival (OS) endpoint set to be completed at a later date.

Intismeran autogene is a truly individualized therapy, crafted for each specific patient. It works by delivering mRNA instructions that guide the immune system to identify and attack up to 34 distinct neoantigen targets present on the patient's tumor. Notably, the core research and development process for this vaccine heavily relies on advanced AI technology. According to previous disclosures from Moderna, the platform utilizes a suite of integrated AI algorithms to read sequencing data from a patient's tumor and blood samples, analyzing genetic mutations to predict which ones are most likely to generate an effective immune response. From this analysis, the system selects up to 34 neoantigens, which then serve as the blueprint for designing and manufacturing a bespoke mRNA drug for that individual.

Moderna's Chief Executive Officer, Stéphane Bancel, hailed the outcome as a "remarkable milestone for mRNA science." The two companies plan to immediately initiate discussions with regulatory authorities regarding marketing authorization applications. If all goes well, the product could potentially receive approval and hit the market as early as 2027. Detailed data from the trial is expected to be presented at upcoming international medical conferences. Dr. Dean Li, President of Merck Research Laboratories, commented in the announcement: "These results further substantiate the potential of personalized cancer treatment. We believe individualized neoantigen therapy has the capacity to redefine the clinical treatment paradigm for patients with completely resected stage IIB-IV melanoma."

Despite the market's enthusiastic response, several financial institutions have adopted a cautious, neutral stance on the true value of these Phase III results. Analysts at Citigroup have indicated that for the results to be considered "positive," the hazard ratio (HR) for recurrence-free survival must be statistically significant and equal to or less than 0.72. A more definitive "clear success" would be an HR of 0.65 or below. A hazard ratio of 0.65 would imply a 35% reduction in the risk of recurrence for the combination therapy compared to the placebo-plus-pembrolizumab arm. In earlier clinical studies, the vaccine demonstrated a five-year recurrence-free survival hazard ratio of 0.51. However, the Citigroup analysts pointed out that because the Phase III patient population is broader and patients may experience recurrence before their customized vaccine is manufactured, the hazard ratio in this Phase III readout is expected to be somewhat less favorable, or "narrower," than the earlier figure.

Adding another layer of perspective, analysts at Jefferies expressed their view last month that a recurrence-free survival hazard ratio falling within the 0.5 to 0.8 range would already "possess clinical value." They also noted that this positive result could provide a significant boost to other ongoing Phase II and III trials evaluating the vaccine across a range of solid tumors. This is not the first time the vaccine has sought regulatory approval. Moderna previously filed for accelerated approval for this cancer vaccine in 2024, but was turned down by the U.S. Food and Drug Administration (FDA). This new positive data may strengthen their position in future regulatory submissions.

Furthermore, Moderna's pioneering mRNA-based influenza vaccine, the world's first of its kind, has just received FDA approval. As a trailblazer in the mRNA field, Moderna's recent corporate strategy has been to diversify its product pipeline, moving beyond its traditional focus on infectious diseases to place greater emphasis on its oncology portfolio. Investors have been eagerly awaiting late-stage clinical data to validate this strategic pivot. This week, it appears they received the key evidence they were hoping for. Following the positive announcement, Moderna's stock price soared to multi-year highs. In regular trading on Wednesday, shares rocketed up 177% to close at $174.38 per share.

The positive news also triggered a significant rally in Asian markets. On the morning of August 20, innovative drug and vaccine sectors on both the A-share and Hong Kong stock exchanges opened sharply higher. A wave of limit-up trading was seen in stocks such as Lummy Pharmaceutical, Walvax Biotechnology, CanSino Biologics, Jiankai Technology, Yuekang Pharmaceutical, SL Pharmaceutical, and Beijing Biosino Biotechnology. Meanwhile, Chongqing Zhifei Biological Products, CSPC Innovation Pharmaceutical, and Shenzhen Kangtai Biological Products all recorded gains of more than 10%.

Analysts believe this trial outcome is crucial for Moderna's growth trajectory and for "rekindling shareholder interest in the company." After generating billions of dollars in revenue from its COVID-19 vaccine, Moderna has been grappling with the pressure of a shrinking vaccine market. While Merck and Moderna have not yet provided a timeline for submitting a marketing application to the FDA, analysts have pointed out that these topline results validate the feasibility of the mRNA technology platform, which is currently being investigated in clinical studies for other cancers, including lung, bladder, and kidney cancer.

The market reaction indicates that Moderna is already "starting to gain a substantial valuation premium in other tumor type indications." However, they also cautioned that this Phase III result primarily serves to "increase the probability of success for trials in other cancers" rather than directly validating the platform's efficacy in those settings. Moderna expects to present the full dataset from the melanoma trial at an upcoming medical conference and may also disclose interim results from its renal cell carcinoma study by the end of this year.

Analysts further suggest that several core questions remain unresolved. "As time goes on, the market will gradually clarify: what is the appropriate valuation range for this stock? How much risk has been released from the non-small cell lung cancer pipeline, and how should that value be calculated? What is the risk profile for the renal cell carcinoma pipeline and its corresponding worth? These are the core fundamental questions Moderna must address moving forward," one analyst stated. In a research note, Citigroup analysts also wrote that Moderna's pipeline "overall remains in a phase requiring data validation," and they believe "broader questions regarding execution and commercialization still remain to be answered."
